Bone marrow fibroblasts in patients with advanced lung cancer BJMBR
Chasseing,N.A.; Hofer,E.; Bordenave,R.H.; Shanley,C.; Rumi,L.S..
In a previous study we demonstrated that the incidence of fibroblast colony-forming units (CFU-F) was very low in bone marrow primary cultures from the majority of untreated advanced non-small lung cancer patients (LCP) compared to normal controls (NC). For this reason, we studied the ability of bone marrow stromal cells to achieve confluence in primary cultures and their proliferative capacity following four continuous subcultures in consecutive untreated LCP and NC. We also evaluated the production of interleukin-1ß (IL-1ß) and prostaglandin E2 (PGE2) by pure fibroblasts. Bone marrow was obtained from 20 LCP and 20 NC. A CFU-F assay was used to investigate the proliferative and confluence capacity. Levels of IL-1ß and PGE2 in conditioned medium (CM) of...

Cellular and matrix interactions during the development of T lymphocytes BJMBR
Owen,J.J.T.; McLoughlin,D.E.; Suniara,R.K.; Jenkinson,E.J..
The thymus contains an extensive extracellular matrix. Although thymocytes express integrins capable of binding to matrix molecules, the functional significance of the matrix for T cell development is uncertain. We have shown that the matrix is associated with thymic fibroblasts which are required for the CD44+ CD25+ stage of double negative (CD4-8-) thymocyte development. The survival of cells at this stage is dependent on IL-7 and we propose that the role of fibroblasts is to present, via the matrix, IL-7 to developing T cells.

Control of the skin scarring response Anais da ABC (AABC)
Ferreira,Lydia M.; Gragnani,Alfredo; Furtado,Fabianne; Hochman,Bernardo.
There comes a time when the understanding of the cutaneous healing process becomes essential due to the need for a precocious tissue repair to reduce the physical, social, and psychological morbidity. Advances in the knowledge on the control of interaction among cells, matrix and growth factors will provide more information on the Regenerative Medicine, an emerging area of research in medical bioengineering. However, considering the dynamism and complexity of the cutaneous healing response, it is fundamental to understand the control mechanism exerted by the interaction and synergism of both systems, cutaneous nervous and central nervous, via hypothalamus hypophysis-adrenal axis, a relevant subject, but hardly ever explored. The present study reviews the...

Effect of amikacin, cephalothin, clindamycin and vancomycin on in vitro fibroblast growth Genet. Mol. Biol.
Souza,Fernanda Timm Seabra; Mello,Alexandre Silva de; Michelin,Kristiane; Coelho,Janice Carneiro.
The effect of four antibiotics (amikacin, clindamycin, cephalothin and vancomycin) was investigated considering that bacterial infection in fibroblasts cultures is a very frequent event. The investigation included the effect of the antibiotics on fibroblast growth and on the activity of the enzyme glucocerebrosidase. The antibiotics were added to the fibroblast cultures and cell growth was evaluated by counting the number of cells and their viability. After cell harvesting, the enzyme activity and content of protein were measured. The results allowed us to conclude that none of the antibiotics affected the cellular number nor the cellular viability. The content of protein decreased when cephalothin and clindamycin were added to the cultures, and...

Effects of plant lectins on in vitro fibroblast proliferation BABT
Sell,Ana Maria; Costa,Celso Paulino da.
Lectins are carbohydrate-binding proteins that have been isolated from various sources and presented a wide spectrum of biological activities. The effects of four lectins, namely, Phaseolus vulgaris phytohemagglutinin, PHA, wheat germ agglutinin, WGA, Artocarpus integrifolia seed lectins, jacalin and artocarpin, on in vitro fibroblasts proliferation were investigated. The lectins did not influence the initial cell adhesion to the plate. PHA and WGA at 10-20 µg/mL concentrations significantly decreased fibroblasts proliferation. At these concentrations, they caused morphological alterations on cells and over 80 µg/mL, promoted cell death. Neither jacalin nor artocarpin significantly affected cell proliferation.

Morphometric and ultrastructural studies of the effect of infrared laser on rabbit temporo-mandibular joint fibroblasts BABT
Uribe,Ricardo Cornejo; Vargas,Fernando Matamala; Mella,Héctor Silva; Onate,Orlando Garrido.
The infrared laser beams has been successfully used in joint lesions treatment due to its analgesic anti inflamatory and healing action, and on a cellular level, inducing an accentuated synthesis of ATP, activating cellular proliferation and collagen synthesis. Samples of normal-control and irradiated tissue from temporo-mandibular joint disc of rabbit, were obtained, with daily doses of 2 joules/cm²during 10 consecutive days. They were prepared for transmission electronic microscopy.Transmission electronic micrographs of normal and irradiated fibroblasts were obtained with final increase of 11.500 X for morphometric studies with the purpose of quantifying the volumetric fractions of the cellular components pertaining to the mentioned cellular types....
